Effects of plant growth regulators on transient expression of foreign gene in Nicotiana benthamiana L. leaves

Abstract Background In the last decades, replicating expression vectors based on plant geminivirus have been widely used for enhancing efficiency of transient expression. By using vector derived from bean yellow dwarf virus and green fluorescent protein as a reporter, we investigated effects α-naphthalene acetic acid, gibberellins 3 , 6-benzyladenine, three common growth regulators, biomass during process in Nicotiana benthamiana L. leaves. Results With increase concentration 6-benzyladenine (from 0.1 to 1.6 mg/L), fresh weight, dry leaf area seedlings increased first then returned levels similar controls (without chemical treatment). The treatment with acid at 0.2 0.4 mg/L can enhance level protein, which peaked was about by 19%, compared controls. Gibberellins 0.1–0.4 25%. However, application led decrease protein. Conclusions appropriate regulators moderate could be beneficial foreign genes Agrobacterium -mediated system plants. Thus, considered exogenous components that are applied production recombinant plant-based systems.
